Lehrinhalt

Physiologic 3D scaffold-based and scaffold-free cell cultivation under dynamic conditions are critical for in vitro engineering of tissue constructs or expansion of functional stem cells.
Dynamic (bioreactor) culture enables the generation of a microenvironment, similar to (human) in vivo conditions. For this, bioreactor systems, as well as specialized incubator systems, are required to apply defined mechanical forces and enable automated nutrient and waste management.

In this course, the students will gain theoretical and practical knowledge about different aspects of 3D culture in bioreactor systems with the aim to establish more relevant culture systems for the generation of engineered tissue constructs.

Topics that are covered in the lectures include:
• Advanced principles of 3D cell cultures
• 3D biomaterials (collagen and ceramic) and Scaffold-free 3D
cultures
• Parameters for optimization of 3D cultivation conditions
• Impact of mechanical forces on the differentiation of stem cells
• Monitoring of dynamic 3D culture systems
• Methods for the generation of 3D structures
• Applications of dynamic 3D cultivation systems
• Approaches for dynamic seeding of cells on scaffolds

o perfusion bioreactors
o stirred tank bioreactors
o vertical wheel bioreactors
o Commercially available 3D cultivation materials and devices

Inhaltliche Voraussetzungen (erwartete Kenntnisse)

Master and PhD students of biotechnology or related subjects (Bachelor students will not be accepted). The course is tailored for students who already are familiar with
standard cell cultivation techniques as well as 3D cell culture (see also “Teilnahmekriterien & Anmeldung”) and want to extend their theoretical and practical knowledge to the specialized field of dynamic 3D cell culture.

Lehrziel

After successful participation in this course, students will be able to
- practically perform dynamic 3D cultivation techniques and operate bioreactor systems.
- describe the basics of physiologic cultivation of cells, tissues, and in vitro models including theoretical background, current research, and applications.
- know and can describe systems for the generation of 3D structures either scaffold-based or scaffold-free, as well as a suitable bioreactor and incubation systems to culture different models.
- explain and evaluate methods used for 3D cell cultivation and analysis and know their purpose and limitations.
- follow and join discussions on this topic.
- present the results both in written and oral form.
