Content

Physiologic 3D scaffold-based and scaffold-free cell cultivation
under dynamic and hypoxic conditions is critical for establishing
relevant in vitro models as an alternative for animal models (3R).
Dynamic cultivation enables automated nutrient and waste
management as well as the application of mechanic or hydrostatic
stress. Therefore, bioreactor systems as well as specialized
incubator systems are required. Furthermore, those systems will be
demonstrated to be used under hypoxic condition mimicking the
physiological oxygen level within the body, below the ambient
oxygen concentration. In this course, the students will gain
knowledge about different aspects of 3D cell cultivation, including
to establish in vivo like conditions for the culture of relevant in
vitro alternatives to animal testing.

Previous knowledge expected

Master and PhD students of biotechnology or related subjects
(Bachelor students will not be accepted).
The course is tailored for students who already are familiar with
standard cell cultivation techniques as well as 3D cell culture (see
also “Teilnahmekriterien & Anmeldung”) and want to extend their
theoretical and practical knowledge to the specialized field of
dynamic 3D cell culture.

Objective (expected results of study and acquired competences)

Students will be able to describe the basics of physiologic
cultivation of cell, tissues and in vitro models including theoretical
background, current research and applications. Moreover, the
students will be aware of replacing animal experiments by more
relevant tissue models cultured under physiologic conditions
resembling the in vivo situation. They know systems for the
generation of 3D structures either scaffold-based and scaffold free,
as well as suitable bioreactor and incubation systems to culture
different models. After the successful participation in the course,
the students can explain and evaluate methods used for 3D cell
cultivation and analysis and know their purpose and limitations.
Furthermore, the students are able to follow and join discussions on
this topic. The students can also practically perform dynamic 3D
cultivation techniques and operate bioreactor systems. The students
can present the results both in written and oral form.
