Content

Protein engineering by rational, semi-rational and random methods is an important method in adapting properties of protein. In this practical course two examples, engineering of an enzyme by a semi-rational approach (site-saturation mutagenesis at a selected position with subsequent screening for improved activity in MTP) as well as a random approach (yeast display of a randomly constructed antibody library, a scFC library, and FACS-based screening of improved thermostability) will be given as examples

Previous knowledge expected

basic knowledge in molecular biology and protein / enzyme chemistry
.

Objective (expected results of study and acquired competences)

knowledge on different methods of protein engineering (rational and random approaches to engineer proteins)
.
