Content

Mutagenesis will be performed on a plasmid containing the gene for a serine protease. After transformation in a suitable expression host (Bacillus subtilis), colonies will be pre-screened on plates with special medium. Selected colonies will be cultivated in liquid medium, and the resulting secretory enzymes will be tested for proteolytic activity and improved thermostability. Mutants with improved properties are selected and sequenced.

Knowledge of microbiology/molecular biology highly is required

Objective (expected results of study and acquired competences)

There is an increasing demand for improved enzymes for industrial application, e.g. improved thermo stability, and altered substrate specificity. The methods of choice for producing this enzymes are directed evolution, site directed mutagenesis, gene shuffling, and a mixture of this methods. Aim of the course is an understanding of the principles of these methods, knowledge about what methods are recommended under what circumstances, and practical competence in applying one method in the lab.
