The seminar will focus on hands on laboratory work and computer-assisted analysis, with some short lecture or presentation material to augment what was covered in the the Lecture "Population genetics and evolutionary theory relevant for the management and protection of aquatic organisms".
Practical work will include DNA extraction, measurement and quantification of DNA/RNA, a PCR and sequence reaction, both fragment (microsatellite) and sequence data analysis, starting with the raw electropherograms through to statistical analysis and interpretation. To provide experience with a range of techniques and questions, some work will be done by the students from the beginning to the end, while other forms of data preparation and analysis will be done with existing data, or data produced by the lecturer or assistant during the course. Emphasis will be given to quality control, troubleshooting and an understanding of the strengths and weaknesses of different types of data. Internet resources (e.g. Genbank) will also be interactively introduced and applied in an analytical context.
- Inhaltliche Voraussetzungen (erwartete Kenntnisse)
Students are expected to have successfully completed the lecture "Population genetics and evolutionary theory relevant for the management and protection of aquatic organisms"
After successful completion of this module, participants are able to:
•Understand protocols and theory for DNA extraction, DNA and tissue preservation, PCR, DNA sequencing and microsatellite typing;
•Apply protocols for DNA extraction, DNA and tissue preservation, PCR, DNA sequencing and microsatellite typing in the laboratory (hands on practice in lab);
•Apply methods for genetic data analysis including introduction to various software programs and internet tools.
