The lecture series will focus on the impact that X-ray crystallography has had in the physical and life sciences. In Part I, we will look at the the intricacies of structure determination and how they allow us to have confidence in a result, or not. In Part II, we will look at how X-ray crystallography has changed our understanding of molecular structure, with an emphasis on the structural biology of proteins.

•Part I:
oThe importance of symmetry in crystal structure determination
oApproaches to solving the phase problem
oResolution & Refinement: The limits of our knowledge

•Part II:
oA history of structure determination, from Nobel Prize to Nobel Prize
oAntibody structure & specificity
oStructure and function of glycosyltransferases

Inhaltliche Voraussetzungen (erwartete Kenntnisse)

Students will be assumed to have a fundamental knowledge of chemistry and biochemistryand an interest in methods of structure determination by single crystal X-ray diffraction. No previous knowledge of X-ray crystallography is required


Students will be familiar with fundamentals and modern developments of structure determination of macromolecules by X-ray crystallography and will learn relevant applications of the technique to the structuralbiology of antibodies and enzymes in particular and macromolecules in general. Students should be able to understand and critique literature on the determinations of structures by these methods.​
