Selection of topics for Upstream processing

- medium/supplements
- cultivation modes and feeding strategies
- up scaling strategies
- cultivation systems for anchorage dependent cells (e.g. stem cells)
- single use equipment and disposables
- examples of different production systems/bioreactors/process modes for biotechnological and biopharmaceutical products incl. modern cell based therapy products

Downstream Processing
•Host cell derived impurities (Protein, DNA, endotoxins, critical impurities)
•Flocculation, Precipitation, Crystallization
•Extraction
•Protein refolding
•Removal of impurities and adventitious agents
•Virus/TSE inactivation strategies
•Continuous purification strategies (SMB, CCE, CCC)
•Purification of antibodies, plasmids, viruses, and virus like particles
•Protein stabilisation

Inhaltliche Voraussetzungen (erwartete Kenntnisse)

Basic chemical engineering knowledge is requested. This includes basics in heat, mass, momentum transfer, and basics in thermodynamics. Solutions of differential equations (ODE solver). Furthermore basic knowledge in bioporocess engineering and cell culture technology as well as cell biology is desirable

Lehrziel

After successful completion of this course, students are able:
-to select suitable medium composition and supplements, choice of production host, select suitable bioreactor, cultivation mode and feeding strategy as well as unit operations for a given biotechnological production problem, such as manufacturing of bioactive compounds/cell based therapeutics.
-to select appropriate quality and in-process control methods and standards for biotechnological manufacturing.
