About the instrument

The Leica TCS SP5 is a laser (point) scanning confocal microscope that enables imaging of transparent, medium thick samples via optical sectioning. Specimens/objects of interest should be fluorescently labelled for image acquisition.

Labelling protocols should be adapted to the existing laser lines that can excite the reporter dye molecules or fluorescent proteins.

Technical details

  • Microscope stand: DM6000, upright configuration
  • Scanning head: SP5 II with Acousto-Optical Beam Splitter (AOBS) and resonant scanner (8 kHz)
  • Stage: motorized, inserts for slides, Petri dishes and microtiter plates
  • Detectors: 2 HyDs, 3 PMTs + TLD
  • Lasers:
    405 nm (cw 50 mW)
    Argon laser (5 lines)
    561 nm
    633 nm

Objectives (default set):

  • HC PL APO CS  10x/0.40  Dry,  WD: 2.2,  DIC,  15506285)
  • HC PL APO CS  20x/0.70 ImmCorr,  WD: 0.26-0.17,  CCorr.  DIC,  (15506326)
  • HCX PL APO CS  40x/0.85  Dry,  WD: 0.21,  CCorr.  (15506295)
  • HCX PL APO CS  63x/1.4  Oil,  WD: 0.1,  (15506188)
  • HCX PL APO CS  63x/1.30 Glyc,   WD: 0.28,  CCorr.  (15506194)
  • HCX  APO L  40x/0.80  Water,  WD: 3.3,  UV - V - I   (15506155)
  • HCX  APO L  63x/0.90  Water,  WD: 2.2,  UV - V - I   (15506148)

Visual observation of the fluorescent signal is possible by illuminating the samples with a metal halide lamp, however, the stand holds no camera for non-confocal image acquisition.
